What L-Tryptophan Is
L-tryptophan is an essential amino acid — essential meaning the body cannot make it, so it has to come from food. It is abundant in meat, eggs and dairy. It also happens to be the raw material the body uses to build serotonin, which is where the entire calming theory comes from.
It appears constantly in pet calming chews, usually near the top of the ingredient list, and in "calm" formulations of dog food.
The Theory, and Why It Is Appealing
The logic is clean: serotonin is central to mood regulation, tryptophan is the precursor to serotonin, therefore more tryptophan should mean more serotonin and a calmer animal. It is the same reasoning behind the folk belief that turkey makes people sleepy.
The problem is a bottleneck. Tryptophan competes with several other large neutral amino acids for the same transporter into the brain, and it is usually outcompeted — especially after a protein-rich meal, which delivers plenty of the competitors alongside it. Supplementing tryptophan does not straightforwardly raise brain serotonin.
What the Evidence Actually Shows
This one has been examined properly, and the answer is worth knowing before you pay for it.
A Veterinary Evidence Knowledge Summary appraised the available trials and concluded that dietary tryptophan supplementation "is thus not enough as a sole treatment to assist in reducing anxiety in anxious adult dogs." Two studies met the inclusion criteria. One found no meaningful effect on anxiety-related behaviour at home. The second found that owners perceived less anxious behaviour — but urine cortisol-to-creatinine ratios showed no physiological difference at all.
That pattern — reported improvement without a matching biological signal — is the classic shape of an expectation effect, and it is exactly why open-label supplement studies should be read carefully.
Going further back, the well-known DeNapoli et al. study in JAVMA examined dietary protein content and tryptophan supplementation against dominance aggression, territorial aggression and hyperactivity in dogs. The findings were narrow rather than sweeping.
The fair reading: tryptophan is not useless, but it is not a standalone answer, and a chew that leans on it as the primary active is leaning on thin ice. We grade the whole shelf in what's actually in calming treats.
How Much to Give
Commonly used amounts sit around 5 to 10 mg per pound of body weight daily, usually divided. Practically that is roughly 100–200 mg daily for a 20 lb dog and 250–500 mg for a 50 lb dog.
One practical detail that follows from the biology above: tryptophan is generally given away from a high-protein meal, because a protein load supplies the competing amino acids that crowd it out at the transporter. A small carbohydrate-containing snack is the usual vehicle.
Judge it over two weeks of consistent use. It is not a same-day tool.
Side Effects and Cautions
The most important caution in this entry is an interaction, not a side effect.
Serotonin syndrome risk. Do not combine tryptophan with serotonergic medication — SSRIs like fluoxetine, trazodone, clomipramine, or an MAOI such as selegiline — without veterinary direction. Too much serotonergic activity at once can cause tremors, agitation, a raised heart rate, high temperature and, in serious cases, collapse. This is a genuine risk, not a theoretical one, and behavioural medication is common in exactly the dogs whose owners buy calming chews.
Stomach upset and mild drowsiness at higher amounts.
Liver or kidney disease — amino acid loads are a vet conversation in these animals.
Pregnant and nursing animals — not studied.
Why "Calming" Diets Use It
Tryptophan is one of the few calming ingredients that shows up in prescription and over-the-counter diets rather than only in supplements, and the reason is the transport problem above. A food formulated for calm can do something a capsule cannot: it can control the ratio of tryptophan to the competing amino acids in the same meal, usually by moderating total protein and adjusting the amino acid profile.
Whether that produces a behavioural result is the same open question — but it is at least a mechanistically coherent attempt, which is more than can be said for adding tryptophan powder on top of a high-protein dinner.
The Cofactors Nobody Mentions
Converting tryptophan into serotonin is not a single step. The pathway needs vitamin B6 as a cofactor, along with adequate iron and niacin status. This is why better calming formulations pair tryptophan with B vitamins rather than including it alone, and it is a reasonable thing to look for on a label.
It also means a tryptophan supplement given to an animal on a complete, balanced diet is unlikely to be correcting a deficiency — the cofactors and the tryptophan are both already present in adequate food. You are attempting to push a system, not fill a gap.
Cats and Horses
Cats are obligate carnivores with a naturally protein-heavy diet, which makes the competition problem above even more pronounced. Tryptophan appears in feline calming products, often with alpha-casozepine. Use a cat-formulated product and see cat safety.
Horses are where tryptophan is most heavily marketed — it is in a large share of equine calming supplements. The evidence there is no stronger than in dogs, and some equine work has found no effect or even mild excitability at low doses. Competition rules on tryptophan vary by governing body; check before a show.
How to Choose a Product
The usual discipline applies and matters more here than most: milligrams of L-tryptophan printed per serving, not buried in a proprietary blend; the L- form named; and third-party testing behind it. Given that the evidence says tryptophan is not sufficient alone, a product that pairs a real amount of it with a real amount of something else is a more sensible buy than one that leads with it.
When to Involve Your Veterinarian
Before starting, if your animal takes any behavioural medication — this is the interaction above and it is the one that can genuinely hurt. Also if the behaviour is panic rather than nerves: self-injury, destruction, escape attempts, refusing food. The American Kennel Club notes that pet supplements are not subject to standardised testing, which is the broader reason to loop a vet in rather than relying on a label.
